Effects of Antioxidant Activities and Hair Loss Prevention of Platycodon grandiflorum Ethanol Extract

Abstract
In this study, we¡¯ve examined the effects of antioxidant activities and hair loss prevention of ethanol extract from Platycodon grandiflorum. We prepared ethanol extract from P. grandiflorum, which is known to have antioxidant activity. Free radicals, which are known to accelerate the aging process, could also inhibit the hair vitality by damaging the membrane of hair follicle. Evaluation of the antioxidant activities based on the 1,1-diphenyl-2-picrylhydrazyl radical and 2,2¡¯-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity revealed that this extract exhibited dose-dependent antioxidant activities. To investigate its ability to prevent hair loss, the effects of the extract on skin regeneration, inhibition of inflammation and cell proliferation were investigated using human skin cells (HaCaT), RAW264.7 cells, and hair follicle dermal papilla cells (HFDPC cells), respectively. In addition, the inhibitory activity of the extract toward dihydrotestosterone (DHT) production was investigated. The assay of RAW264.7 cells revealed the extract exerted an inflammation inhibitory effect by suppressing the production of nitric oxide and prostaglandin E2. The HFDPC cell proliferation assay revealed that the extract promoted cell proliferation after 24 h treatment with 1, 10, and 100 ¥ìg/mL. Finally, the extract inhibited the DHT production at 1, 10, 50, and 100 ¥ìg/mL. Taken together, these results confirm the value of ethanol extract from P. grandiflorum as a useful raw material for hair loss prevention products.
